چکیده

Background & Objective: The current approaches to reduce the risk of colorectal carcinoma are through the detection and removal of the precursor lesion” adenomatous polyps”. The study was conducted to evaluate the immunohistochemical expression of p53 and bcl-2 in colorectal adenomas and carcinomas. Patients and Methods: A total of 86 cases, 33 colorectal adenomas, 33 colorectal adenocarcinomas and 20 samples of non -tumerous colonic tissue as control, were included in this retrospective study. Sections were stained immunohistochemically for p53 and bcl-2. Scoring was performed using Digimizer software. Data were analyzed using SPSS program (Statistical Package for Social Sciences) version 16 and Microsoft Office Excel 2007. Results: The frequency of p53 positive cases was significantly higher in carcinoma than adenoma while the frequency of bcl-2 positive cases was significantly higher in adenoma than carcinoma. p53 expression was significantly higher in large sized adenomas, villous configuration, severe dysplasia, and multiple lesions. Bcl-2 expression showed significantly correlated with adenomas of small size, solitary, tubular, and mild dysplasia. There was a significant correlation between bcl-2 expression and non-mucinous carcinoma and a negative correlation with tumor size. There was an inverse relationship between bcl-2 and p53 expression in both colorectal adenomas and carcinomas. Conclusion: p53 overexpression is a late event in colorectal tumorigenesis. bcl-2 may play an important role in the early stage of adenoma-carcinoma sequence.

Histopathological Evaluation and Analysis of Immunohistochemical Expression of Bcl-2 Oncoprotein in Colorectal Carcinoma

Abstract Introduction: Colorectal cancer is the third most prevalent malignancy with high mortality rate, necessitating markers that predict survival and guide the treatment. Previous studies have examined the immunohistochemical expression of Bcl-2, an apoptotic marker, in colorectal carcinoma, but results have been contradictory. Objectives: To evaluate the histopathological featu...

Genes mediating programmed cell death: an immunohistochemical study of bcl-2, c-myc and p53 expression in colorectal neoplasia.

Aims-To describe the expression of three genes involved in the regulation of cell proliferation and programmed cell death (apoptosis) in normal, dysplastic and malignant large bowel epithelium, and to relate any alterations to important biological and clinical variables.Methods-Immunohistochemistry was used to assess bcl-2, c-myc and p53 gene expression in 70 colorectal carcinomas, 36 adenomas ...

Bcl-2 expression in colorectal tumours. Correlation with p53, mdm-2, Rb proteins and proliferation indices.

Immunostaining for bcl-2 protein was performed in 27 colorectal adenomas and 108 colorectal adenocarcinomas. The aim of the study was to determine bcl-2 expression in correlation with p53, mdm-2 and Rb expression, with proliferation indices (Ki-67-LI, PCNA-LI) as well as with conventional clinicopathological variables. A higher proportion of adenomas (30.8%) than carcinomas (16.7%) expressed bc...

Expression of p53, p21/waf, bcl-2, bax, Rb and Ki67 proteins in colorectal adenocarcinomas.

This study investigated the combined immunoexpression of p53, p21, bcl-2, bax, Rb and Ki67 proteins in colorectal adenocarcinomas and correlated expression patterns with tumour stage and grade. Paraffin sections from 98 cases of colorectal adenocarcinomas were stained by immunohistochemistry for p53, p21, bcl-2, bax, Rb and MIB-1 (Ki67) proteins. In addition, 12 cases of colorectal adenomas and...
